چکیده

Clone A colon carcinoma cells develop fanshaped lamellae and exhibit random migration when plated on laminin, processes that depend on the ligation of the a 6 b 4 integrin. Here, we report that expression of a dominant negative RhoA (N19RhoA) in clone A cells inhibited a 6 b 4-dependent membrane ruffling, lamellae formation, and migration. In contrast, expression of a dominant negative Rac (N17Rac1) had no effect on these processes. Using the Rhotekin binding assay to assess RhoA activation, we observed that engagement of a 6 b 4 by either antibody-mediated clustering or laminin attachment resulted in a twoto threefold increase in RhoA activation, compared with cells maintained in suspension or plated on collagen. Antibody-mediated clustering of b 1 integrins, however, actually suppressed Rho A activation. The a 6 b 4-mediated interaction of clone A cells with laminin promoted the translocation of RhoA from the cytosol to membrane ruffles at the edges of lamellae and promoted its colocalization with b 1 integrins, as assessed by immunofluorescence microscopy. In addition, RhoA translocation was blocked by inhibiting phosphodiesterase activity and enhanced by inhibiting the activity of cAMP-dependent protein kinase. Together, these results establish a specific integrin-mediated pathway of RhoA activation that is regulated by cAMP and that functions in lamellae forma-

A novel cAMP-dependent pathway activates neuronal integrin function in retinal neurons.

Retinal neurons lose the ability to attach to and extend neurites on substrata of laminin-1 (LN-1) during late embryogenesis, in a time frame that corresponds to target innervation. Although this developmental loss correlates with a modest downregulation of integrin expression, we have shown previously that these neurons use the same laminin-binding integrins for outgrowth on other laminin isof...

Cyclic AMP induces integrin-mediated cell adhesion through Epac and Rap1 upon stimulation of the β2-adrenergic receptor

cAMP controls many cellular processes mainly through the activation of protein kinase A (PKA). However, more recently PKA-independent pathways have been established through the exchange protein directly activated by cAMP (Epac), a guanine nucleotide exchange factor for the small GTPases Rap1 and Rap2. In this report, we show that cAMP can induce integrin-mediated cell adhesion through Epac and ...

Cyclic AMP regulates formation of mammary epithelial acini in vitro

Epithelial cells form tubular and acinar structures notable for a hollow lumen. In three-dimensional culture utilizing MCF10A mammary epithelial cells, acini form due to integrin-dependent polarization and survival of cells contacting extracellular matrix (ECM), and the apoptosis of inner cells of acini lacking contact with the ECM. In this paper, we report that cyclic AMP (cAMP)-dependent prot...
